The Effect of Negative Environment Controlled Rearing System(ECRS) to Prevention of Hantaan Virus Horizontal Transmission in the Apodemus aqrarius and Hamsters

Abstract
Hemorrhagic fever with renal syndrome(HFRS) is a disease caused by a viruses beloning to a Hantavirus Genus of the Family Bunyaviridae. These viruses are distributed throughout the world among various rodent species. Large quantities of Hantaan virus are excreted in the urine, saliva and feces of infected mice, Apodemus agrarius. Many investigators and animal breeders have been affected from animal study with infectious agents. Negative-type Environment controlled Rearing System(ECRS) is absolutely necessary to such artificial infection research. This study performed to assess preventive ability of horizontal transmission from Hantaan virus infected Apodemus agrarius and hamsters to noninfected Apodemus agrarius and hamsters in negative ECRS that manufactured in Korea. Furthermore, we investigated the bio-safety of negative ECRS whether horizontal transmission outbreak to researchers and breeders occur or not.
In usual ECRS, artificially inoculated Apodemus agrarius and hamsters have antibody titers against Hantaan virus. Lower and same floor left groups of inoculated Apodemus agrarius and Lower, same floor left and right groups of inoculated hamsters were antibody positive, respectively. With indirect fluoresence antibody(IFA) method and RT-PCR for antigen test of Hantaan virus in lung and kidney, there were positive in lower and same floor left and right groups of inoculated Apodemus agrarius and in lower group of inoculated hamster.
In improved ECRS, artificially inoculated Apodemus agrarius and hamsters have antibody titers against Hantaan virus. Lower shelf group from inoculated Apodemus agrarius was positive but all groups around inoculated hamsters were negative. No Hantaan virus antigen was detected in lung and kidney using IFA method and RT-PCR.
It was considered that improved ECRS have more biosafe effect than usual ECRS in air-borne viral infection researches. In addition, trends of ECRS development should be improved in circumstance equipments such as regularity and balance of air flow, completeness of shelter system, miniaturization, weight reduction and noise diminution of blower, convenience of opening and shutting.
